Throne-Holst has a master’s degree in public administration and international affairs. She has previously worked at the U.N. Department of Peacekeeping. In 2008, Throne-Holst became a member of the Southhampton Town Board. After serving as a board member for two years, she won election to the position of town supervisor. She was re-elected to the position two more times. Throne-Holst chose not to run for re-election as town supervisor in 2015, opting to run for Congress instead.
